Axl, also known as Ufo and Ark, is a single-pass type I transmembrane glycoprotein in the TAM receptor tyrosine kinase (RTK) family. Like other family members Tyro3 and Mer, Axl contains two immunoglobulinlike (Ig) domains and two fibronectin type III domains in the extracellular region and a cytoplasmic tyrosine kinase domain. The TAM receptors are involved in regulation of immune response, cell survival and migration as well as tumorigenesis and metastasis. Axl binds to its ligand Gas6 (growth arrest specific protein 6), which triggers tyrosine auto-phosphorylation and activation of Axl. Axl induces a wide range of activities including platelet aggregation and thrombus formation, macrophage and dendritic cell phagocytosis, NK cell development, and angiogenesis. Axl is upregulated in various disease states, such as in multiple sclerosis (MS) lesions. Axl contributes to vascular remodeling and inflammatory cell infiltration. Axl acts as a cellular entry receptor for Gas6opsonized lentiviruses. Axl also contributes to tumor cell survival, migration, invasion, metastasis and chemosensitivity. Axl is highly expressed in solid cancers and mediates multiple oncogenic phenotypes. Axl expression correlates with poor prognosis in several cancers. A soluble form of the Axl ectodomain, which is shed by proteolytic cleavage, retains the ability to bind Gas6 and blocks its signaling, showing the therapeutic potential in the inhibition of metastatic tumor progression.

Construct Details: Full length human AXL/UFO gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.
