We offer bundled stable cell line generation service, “Gene-to-Cell-line”, a one-stop shop solution with standardized workflow and reduced turnaround time. Bundle discount is included when you order the target gene expression construct and custom made stable cell lines from G&P Biosciences at the same time. We can synthesize and subcloneyour gene of interest into a suitable expression vector with desired selection marker. We can introduce the DNA construct into the host cells and select the stable pools/clones that express your gene of interest at desirable levels. We can help you validate the genomic integration by genomic PCR and the expression using ELISA, flow cytometry, Western blot, or other cell-based assay of your interest. We can also use established protocols to assess cell proliferation rates and expression stabilities for your custom-made cell lines.

Example #1
The client requested us to generate a stable mouse fibroblast cells expressing a gene of interest (GOI) fused with GFP. In just 16 days, we were able to deliver the stable cells (pools) using our unique gene delivery and cell selection methods. Fluorescence microscopy confirmed GOI’s surface expression (see the image below on the left) and subsequently the stable cells were employed by the client to develop a successful cell-based screening assay.
Example #2
The client requested to produce two independent stable clones of human cancer cells that constitutively and uniformly overexpress a cell surface receptor. In just 6 weeks, we were able to deliver the two stable clones after screening 12 independent single-cell lineages. Flow cytometry analysis with PE-labeled monoclonal antibody (mAb-PE) against this surface receptor confirms the high and uniform surface expression in comparison with the parental cells (see the flow data below for one of the stable clones, #8). The client further used the the stable clones to establish a tumor animal model for in vivo efficacy studies.
