Description

TAJ (Toxicity and JNK inducer), also known as TRADE and TROY, is a single-pass type I membrane glycoprotein in the tumor necrosis factor receptor superfamily (TNFRSF19). TAJ contains 3 TNFR cysteine-rich repeats in the extracellular region but lacks a cytoplasmic death domain (DD). TAJ is widely expressed in the embryo, but shows restricted expression in adult. TAJ is detected in embryonic brain and epithelium, and at lower levels in adult heart, lung and liver. In the adult, TROY is found in the nervous system associated with dorsal root and retinal ganglia neurons, astrocytes, and microglia, and in hair follicles. TAJ has been shown to interact with TRAF family members to activate JNK signaling pathway when overexpressed in cells. TAJ is capable of inducing apoptosis by a caspase-independent mechanism, and it is thought to play an essential role in embryonic development. TAJ may be a component of the Nogo­A receptor that mediates axon repulsion initiated by one of five myelin­associated inhibitory factors. In the hair follicle, TROY binds Lymphotoxin-α and acts with EDA to drive secondary hair follicle development. TAJ has also been found on melanoma cells, not normal melanocytes.  TAJ has thus been identified as a biomarker for melanoma and represents a promising cell surface target for cell and antibody-based immunotherapies.

Construct Details: Full length human TAJ gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
