Description

LFA3 (lymphocyte function-associated antigen 3), also known as Ag3 and CD58, is single-pass type I transmembrane or GPI-anchored glycoprotein belonging to the immunoglobulin superfamily (IgSF). LFA3 contains 1 Ig-like C2-type domain in its extracellular region. LFA3 is primarily expressed on Antigen Presenting Cells (APCs), particularly macrophages.  LFA3 is the ligand of the T-lymphocyte CD2 glycoprotein and functions in adhesion and activation of T lymphocytes. This interaction is important in mediating thymocyte interactions with thymic epithelial cells, antigen-independent and antigen-dependent interactions of T-lymphocytes with target cells. It is important in strengthening the adhesion between the T cells and Professional APCs. This adhesion occurs as part of the transitory initial encounters between T cells and APCs before T cell activation, when T cells are roaming the lymph nodes looking at the surface of APCs for peptide:MHC complexes that the T-cell receptors are reactive to. In addition, the LFA-3/CD2 interaction may prime response by both the CD2+ and LFA3+ cells and is important for the T-lymphocyte rosetting with erythrocytes. Polymorphisms in the LFA3 gene are associated with increased risk for multiple sclerosis.

Construct Details: Full length human LFA3 gene (encoding UniProt #P19256 isoform 1) is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

 

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter+/- a selection marker, see the vector map above)

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
