c-Myc is a proto-oncogenic transcription factor that regulates transcription of target genes important for cell cycle progression, apoptosis and cellular transformation. c-Myc binds DNA in a non-specific manner, yet also specifically recognizes the core sequence 5'-CAC[GA]TG-3' to activates the transcription of target genes. It contains a bHLH (basic helix-loop-helix) and a leucine-zipper (LZ) domain. Efficient DNA binding requires dimerization with another bHLH protein MAX. Mutations, overexpression, rearrangement, and translocation of c-Myc is implicated in the etiology of a variety of hematopoietic tumors, leukemias and lymphomas, such as Burkitt lymphoma. OCT4, SOX2, c-Myc and KLF4 are the four reprogramming factors that together are sufficient to reprogram differentiated cells to an embryonic-like state termed iPS (induced pluripotent stem) cells. iPS cells exhibit the morphology and growth properties of ES cells and express ES cell marker genes. They hold great promise in research involving induced pluripotency, an emerging and promising field of regenerative medicine.

Construct Details: Full length human c-Myc/Myc gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple selection markers are available.
