Oct-4 (Cctamer-binding transcription factor 4) also known as Oct-3 and POU5F1 (POU domain, class 5, transcription factor 1) is a homeodomain transcription factor of the POU family class-5 subfamily. Oct-4 contains 1 homeobox DNA-binding domain and 1 POU-specific domain.  Oct-4 recognizes the octamer core DNA sequence (5'-ATTTGCAT-3'). It forms a trimeric complex with SOX2 on DNA and controls the expression of a number of genes involved in embryonic development. Oct-4 is involved in the self-renewal of undifferentiated embryonic stem cells. It is frequently used as a marker for undifferentiated cells. Oct-4 expression is closely regulated; too much or too little will cause differentiation of the cells. Oct-4 has been implicated in tumorigenesis of adult germ cells. Ectopic expression of Oct-4 in adult mice has been found to cause the formation of dysplastic lesions of the skin and intestine. The intestinal dysplasia resulted from an increase in progenitor cell population and the upregulation of β-catenin transcription through the inhibition of cellular differentiation. Aberrant expression of the oct-4 gene in adult tissues is associated with tumorigenesis. It is found in a translocation with the Ewing's sarcoma gene on chromosome 21, which also leads to tumor formation. OCT4, SOX2, c-Myc and KLF4 are the 4 reprogramming factors that together are sufficient to reprogram differentiated cells to an embryonic-like state termed iPS (induced pluripotent stem) cells. iPS cells exhibit the morphology and growth properties of ES cells and express ES cell marker genes.

Construct Details: Full length mouse Oct4 gene (Uniprot #P20263) is subcloned into the Lentiviral expression vector pLTE with an upstream EF1α promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple selection markers are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
