OPG (Osteoprotegerin), also known as OCIF (osteoclastogenesis inhibitory factor), is a secreted 55 - 60 kDa protein of the TNF-receptor superfamily (TNFRSF11B).  Mature OPG is a 380 amino acid soluble protein consisting of 4 TNFR cysteine-rich repeats, 2 death domains (DD) and 1 heparin-binding region. The cysteine-rich repeats are essential for ligand interaction, while a cysteine at the C-terminus mediates homodimerization.  OPG is widely expressed and constitutively released by mesenchymal stem cells, fibroblasts and endothelial cells. OPG is an osteoblast-secreted decoy receptor that functions as a negative regulator of bone resorption. OPG specifically binds to its ligand, OPG ligand or RANKL (receptor activator of NF-κB ligand), both of which are key extracellular regulators of osteoclast development. OPG and RANKL may also play a role in lymph-node organogenesis and vascular calcification. OPG regulates bone density through inhibiting the activation of osteoclasts and promoting osteoclast apoptosis.  Bone homeostasis may depend on the local RANKL to OPG ratio. OPG also act as a decoy receptor for TRAIL (TNF-related apoptosis-inducing ligand). TRAIL binding blocks the inhibition of osteoclastogenesis. In addition TRAIL decreases the release of OPG from cells that express it, while OPG inhibits TRAIL-induced apoptosis. Binding of OPG by syndecan-1 heparin sulfates on multiple myeloma cells also results in OPG internalization and degradation, contributing to bone loss. OPG deficiency can cause juvenile Paget’s disease (JPD) in humans, and insufficient OPG can produce osteoporosis and vascular calcification in animals and humans.

Construct Details: The recombinant human OPG-Fc fusion is expressed as a 608 amino acid protein consisting of Glu22 - Leu401 region of OPG (UniProt accession #O00300) and a C-terminal Fc from human IgG1, which exists as a dimer under non-reducing conditions (see the gel image inserted above).

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 3 months. The undiluted protein can be stored at 4°C for no more than 2 weeks.

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
