Description

Tyrosine-protein kinase TYRO3, also known as BYK, DTK, RSE, SKY and TIF, is a single-pass type I transmembrane glycoprotein in the AXL/UFO subfamily of the Receptor Tyrosine Kinase (RTK) family. Like other family members AXL/UFO and MER, TYRO3 contains two immunoglobulinlike (Ig) domains and two fibronectin type III domains in the extracellular domain (ECD) and a cytoplasmic tyrosine kinase domain tail. The AXL/UFO family receptors are involved in regulation of immune responses, cell survival and migration as well as tumorigenesis and metastasis. By binding to its ligand, TULP1 or GAS6, TYRO3 transduces signals from the extracellular matrix into the cytoplasm.  Ligand binding induces dimerization and autophosphorylation of TYRO3 on its cytoplasmic tail that provides docking sites for downstream signaling molecules. Following surface activation, TYRO3 enhances PI3-kinase activity and activates the AKT survival pathway, leading to nuclear translocation and activation of NF-kB. TYRO3 signaling is involved in many biological processes such as neuron protection from excitotoxic injury, platelet aggregation and cytoskeleton reorganization. It also plays an important role in inhibition of Toll-like receptors (TLRs)-mediated innate immune response.  TYRO3 acts as a receptor for various virus, including lassa virus, lymphocytic choriomeningitis virus and ebolavirus
