PDGFRA (platelet­derived growth factor receptor alpha), also known as CD140a and PDGFR2, is a single-pass type I transmembrane glycoprotein in the CSF-1/PDGF receptor subfamily of the receptor tyrosine kinases (RTK) family. Like other family members, PDGFRA contains 5 immunoglobulin­like (Ig) domains in the extracellular region and a split tyrosine kinase domain in the intracellular region. PDGFRA and its closely related homolog PDGFRB can form homo­ or hetero­dimeric receptors when engaged by disulfide­linked dimers of the PDGF family of growth factors, including PDGF-A, B, C or D, or the heterodimer PDGF­AB that is mainly found in human platelets. PDGFRA is expressed in oligodendrocyte progenitor cells and mesothelial cell, and binds all three ligand isoforms PDGF-AA, PDGF-BB and PDGF-AB with high affinity. PDGFRA plays an essential role in regulating proliferation, chemotaxis and migration of mesangial cells.  Abnormal PDGFRA signaling in humans may result in spina bifida or cleft palate-type malformations.  PDGFRA may also be a critical mediator of signaling in testis organogenesis and Leydig cell differentiation, and in kidney development. PDGFRA is involved in tumor angiogenesis and maintenance of the tumor microenvironment.  PDGFRA has been implicated in development and metastasis of liver cancer. PDGFRA is implicated in gliomas and fibrotic disorders of lung, heart and skin (scleroderma). PDGFRA gene amplification may be the underlying genetic mechanism driving PDGFRA overexpression in a portion of gliomas.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 3 months. The undiluted protein can be stored at 4°C for no more than 2 weeks.

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
