LYPD8 (Ly6/PLAUR domain-containing protein 8) is a glycosyl-phosphatidylinositol (GPI)-linked cell surface glycoprotein. A variety of GPI-linked surface glycoproteins are composed of one or more copies of a conserved domain of ~100 amino-acid residues termed u-PAR/Ly-6 motif that was originally found in u-PAR (Urokinase plasminogen activator surface receptor) and Ly-6/CD59. The u-PAR/Ly-6 (UL) motif has 10 conserved cysteines with a defined disulfide-bonding pattern. LYPD8 contains 1 copy of u-PAR/Ly6 motifs in the extracellular domain (ECD). The GPI-anchor of LYPD8 can be cleaved, leading to secretion into the colonic lumen. LYPD8 also belongs to the CNF-like-inhibitor family with phospholipase inhibitor activity. LYPD8 is expressed in the large intestine, preferentially on the epithelial layer exposed to the lumen. LYPD8 is required to prevent invasion of Gram-negative bacteria in the inner mucus layer of the colon epithelium, a portion of the large intestine which is free of commensal microbiota. LYPD8 prevents invasion of flagellated microbiota by binding to the flagellum of bacteria, such as P.mirabilis, thereby inhibiting bacterial motility in the intestinal lumen. Segregation of intestinal bacteria and epithelial cells in the colon is required to preserve intestinal homeostasis.

Construct Details: Full length human LYPD8 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
