Description

TPOR (thrombopoietin receptor) or MPL (myeloproliferative leukemia protein) is a single pass type I transmembrane glycoprotein of the type I cytokine receptor family (type 1 subfamily). TPOR is the human homolog of oncogene, v-mpl, which was originally identified from the murine myeloproliferative leukemia virus and capable of immortalize bone marrow hematopoietic cells from different lineages. TPOR is the receptor for thrombopoietin that is the major regulator of megakaryocytopoiesis and platelet formation. TPOR consists of a 466 amino acid (aa) extracellular region with 2 fibronectin type III (FN3) domains and a juxtamembrane WSXWS motif, a transmembrane domain, and a 122 aa cytoplasmic region with a Box 1 motif. The WSXWS motif may be necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding.  The box 1 motif is required for JAK interaction/activation. TPOR deficient mice were severely thrombocytopenic. Upon binding of thrombopoietin, TPOR is dimerized and the JAK family of non-receptor tyrosine kinases, as well as the STAT family, the MAPK family, the adaptor protein Shc and the receptors themselves become tyrosine phosphorylated. Mutations affecting the TPOR gene are associated with congenital amegakaryocytic thrombocytopenia (CAMT) and thrombocythemia 2 (THCYT2).

Construct Details: Full length human TPOR gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
