CD138, also known as SDC1 (syndecan 1), is a transmembrane (type I) heparan sulfate proteoglycan of the syndecan proteoglycan family. The syndecans mediate cell adhesion, signaling, and cytoskeletal organization and syndecan receptors are required for internalization of the HIV-1 tat protein. CD138 is a 310 amino acid (aa) single pass type I transmembrane protein with an N-terminal signal peptide, a 232-aa extracellular domain (ECD) with 6 potential glycosylation sites, a transmembrane domain (TM), and a short 35-aa cytoplasmic tail. CD138 is a cell surface proteoglycan with both heparan sulfate and chondroitin sulfate. CD138 also regulates exosome biogenesis. CD138 shedding is enhanced by a number of factors such as heparanase, thrombin, PMA or EGF, and also by stress and wound healing. CD138 may play a role in cell proliferation, cell migration and cell-matrix interactions via its receptor for extracellular matrix proteins. Altered syndecan-1 expression has been detected in several different tumor types.

Construct Details: Full length human CD138/SDC1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
