Nectin3 or CD113 is a single pass type I transmembrane glycoprotein that belongs to the Nectin family of the Immunoglobulin superfamily (IgSF). Most Nectin family members function as cell adhesion molecules (CAMs) located on the cell surface and involved with the binding with other cells or with the extracellular matrix (ECM). Nectin3 contains 1 Ig-like V-type domain and 2 Ig-like C2-type domains in the extracellular region that promotes cell-cell contacts by forming homophilic or heterophilic trans-dimers. Heterophilic interactions have been detected with NECTIN1, NECTIN2, PVR, IGSF4B/Necl-1 and with IGSF4. Nectin3 also interacts with with afadin, a filamentous actin-binding protein involved in the regulation of directional motility, cell proliferation and survival. Nectin3 may be involved in the formation of cell-cell junctions, including adherens junctions and synapses. Nectin3 induces endocytosis-mediated down-regulation of PVR from the cell surface, resulting in reduction of cell movement and proliferation. Nectin3 plays a role in the morphology of the ciliary body and in ocular development involving the ciliary body. Mutations in the Nectin3 gene are believed to result in congenital ocular defects.

Construct Details: Full length human NECTIN3 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter -/+ an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.
