Nectin1 or CD111 is a single pass type I transmembrane glycoprotein that belongs to the Nectin family of the Immunoglobulin superfamily (IgSF). Most Nectin family members function as cell adhesion molecules (CAMs) located on the cell surface and involved with the binding with other cells or with the extracellular matrix (ECM). Nectin1 contains 1 Ig-like V-type domain and 2 Ig-like C2-type domains in the extracellular region that promotes cell-cell contacts by forming homophilic or heterophilic trans-dimers. Heterophilic interactions have been detected between NECTIN1 and NECTIN3 and between NECTIN1 and NECTIN4. Ig-like C2-type 2 mediates neurite outgrowth through binding, induction of phosphorylation, and activation of FGFR1/2/3. Nectin1 plays a role in the organization of adherens junctions and tight junctions in epithelial and endothelial cells. Nectin1 is a calcium(2+)-independent cell-cell adhesion molecule that acts as a receptor for glycoprotein D (gD) of herpes simplex viruses 1 and 2 (HSV-1, HSV-2), and pseudorabies virus (PRV) and mediates viral entry into epithelial and neuronal cells. Mutations in the Nectin1 gene cause cleft lip and palate/ectodermal dysplasia 1 syndrome (CLPED1) as well as non-syndromic cleft lip with or without cleft palate (CL/P).

Construct Details: Full length human NECTIN1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter -/+ an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.
