Description

CD34 is a single­pass, type I transmembrane protein belonging to the CD34 family that includes Podocalyxin, and Endoglycan. CD34 is a well-known hematopoietic progenitor cell antigen. It is a sialomucin­like molecule that is heavily glycosylated with 9 potential N-linked glycosylation sites in the extracellular domain.  In the cytoplasmic region, CD34 contains serine residues that can be phosphorylated by PKC (protein kinase C), resulting in the recruitment of the hematopoietic adapter proteins and the activation of CD34 signaling pathways. CD34 is expressed on primitive hematopoietic stem cells and down-regulated as they differentiate into mature cells. The precise function of CD34 remains unclear.  The pattern of CD34 expression suggests that it plays an important role in hematopoiesis. CD34 is found on multipotent precursors, bone marrow stromal cells, embryonic fibroblasts, vascular endothelia, mesenchymal stem cells, and some tumor cell lines. CD34 is involved in the adhesion of stem cells to the bone marrow extracellular matrix or to stromal cells. It can act as a scaffold for the attachment of lineage specific glycans, allowing stem cells to bind to lectins expressed by stromal cells or other marrow components. CD34 is overexpressed in many types of tumors and implicated in leukemogenesis.

Construct Details: Expressed as the combination of a heavy chain (HC) containing VH from anti-CD34 mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-CD34 mAb and CL of human kappa light chain.  Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition.

 

Source: Mammalian cells stably expressing anti-CD34 heavy chain and light chain (in human IgG1k format) and growing in chemical-defined media with no animal components.

Formulation: Supplied at 1.0 mg/ml in sterile PBS pH7.4 without any carrier protein or preservative (the concentration determined by UV absorption with an A280nm of 1.0 for 0.73 mg/ml of IgG and verified by SDS-PAGE and Coomassie blue staining)

 

Endotoxin Level: <0.1 EU per 1 μg of purified recombinant protein determined by the LAL method

 

Biological Activity: Binds the immobilized CD34 ECD protein (SKU#FCL1678) with high affinity (KD < 10 nM) by ELISA (see Technical Data).  Has the same binding specificity as anti-CD34 monoclonal antibodies, rabbit IgG (SKU#MAB1713) and mouse IgG2a (SKU#MAB1678).  Recognizes cell surface CTLA4 by flow cytometry/FACS, IHC staining and fluorescent microscopy.

Technical Data

 

Recombinant CD34 ECD protein (SKU#FCL1678) was immobilized at 100 ng/well on a 96-well NuncMaxisorp plate for an ELISA with serially diluted anti-CD34 monoclonal antibody (SKU#MAB0820). Anti-CD3e (SKU#MAB0818) monoclonal antibody was used as a negative control to demonstrate the specificity of CD34 binding under the experimental conditions.  The binding curve was fit and apparent KD was calculated for anti-CD34: CD34 binding to be 3.0 ± 0.4 nM

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 3 months. The undiluted protein can be stored at 4°C for no more than 2 weeks.

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
