Description

CRLF2 (cytokine receptor-like factor 2) is is also known as the receptor for thymic stromal lymphopoietin (TSLP). CRLF2/TSLPR is a single pass, type I transmembrane glycoprotein of the type I cytokine receptor family, type 5 subfamily.  It consists of a extracellular region with 1 fibronectin type III (FN3) domains and a juxtamembrane WSXWS motif, a transmembrane domain, and a cytoplasmic tail with the box 1 motif.  The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding, while the box 1 motif is required for JAK interaction/activation. CD127/IL7R is another component of the receptor complex for TSLP. The TSLP receptor complex is a heterodimer of TSLPR/CRLF2 and CD127/IL7R, which is capable of stimulating cell proliferation through activation of STAT3, STAT5 and JAK2 pathways. CRLF2/TSLPR is implicated in the development of the hematopoietic system.  It is primarily expressed in dendrites and monocytes and weakly expressed in T-cells.  It is up-regulated in activated peripheral monocytes and THP-1 cells. Rearrangement of this gene with immunoglobulin heavy chain gene (IGH) on chromosome 14, or with P2Y purinoceptor 8 gene (P2RY8) on the same X or Y chromosomes is associated with B-progenitor acute lymphoblastic leukemia (ALL) and Down syndrome ALL.

Construct Details: Full length human TSLPR/CRLF2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
