Description

CRHR1 or CRFR1 is a 7 transmembrane receptor belonging to the secretin receptor family, which is also known as family B or 2 of the G-protein-coupled receptors (GPCRs). Many members of this family are receptors for peptide hormones from the glucagon hormone family and regulate the activation of two major signal transduction pathways: adenylyl cyclase (cAMP) and phosphatidylinositol-calcium pathways. CRHR1 binds neuropeptides of the corticotropin releasing hormone (CRH) family that are major regulators of the hypothalamic-pituitary-adrenal pathway. It has high affinity for CRH and UCN (urocortin). Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and down-stream effectors, such as adenylate cyclase. CRHR1 is essential for the activation of signal transduction pathways that regulate diverse physiological processes including stress, reproduction, immune response and obesity. CRHR1 is equired for normal embryonic development of the adrenal gland and for normal hormonal responses to stress. It also plays a role in the response to anxiogenic stimuli.

Construct Details: Full length human CRHR1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
