Description

LINGO1 (Leucine-rich repeat and immunoglobulin-like domain-containing nogo receptor-interacting protein 1) is also known as LERN1 (Leucine-rich repeat neuronal protein 1) and LRRN6A (Leucine-rich repeat neuronal protein 6A). LINGO1 is a single-pass type I transmembrane protein with a 520 amino acid (aa) extracellular region that contains 11 LRR (leucine-rich) repeats and 1 Ig (immunoglobulin)-like C2-type domain. LINGO1 is expressed exclusively in the central nervous system (CNS) with the hghest level in the in amygdala, hippocampus, thalamus and cerebral cortex. In the rest of the brain a basal expression can be detected. LINGO1 is up-regulated in substantia nigra neurons from Parkinson disease patients. LINGO1 is thought to be a functional component of the Nogo receptor signaling complex (RTN4R/NGFR) in RhoA activation.  The receptor complex is responsible for negative regulation of axonal regeneration by myelin-associated factors. LINGO1 may exist as a homotetramer and form a ternary complex with RTN4R/NGFR and RTN4R/TNFRSF19. The intracellular domain of LINGO1 interacts with MYT1L for signal transduction. LINGO1 is also an important negative regulator of oligodentrocyte differentiation and axonal myelination. Lingo1 acts in conjunction with RTN4 and RTN4R in regulating neuronal precursor cell motility during cortical development.  Inhibition of LINGO1 enhances survival, structure, and function of dopaminergic neurons in Parkinson's disease models.

Construct Details: Full length human LINGO1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
