Description

NPR1 (natriuretic peptide receptor 1), also known as GC-A, ANPRA and GUCY2A, is a single pass type I transmembrane glycoprotein that belongs to the adenylyl cyclase class-4/guanylyl cyclase family. Guanylyl cyclases catalyze the production of cGMP from GTP and exist as soluble and membrane forms.  The membrane guanylyl cyclases, often termed guanylyl cyclases A through F, are a family of cell-surface receptors with a similar topographic structure: an extracellular domain (ECD) for ligand-binding, a transmembrane (TM) domain, and an intracellular region that contains a protein kinase-like domain, a helical hinge region involved in oligomerization, and a carboxyl-terminal guanylyl cyclase catalytic domain. NPR1 is a membrane-bound guanylate cyclase that serves as the receptor for both atrial and brain natriuretic peptides (ANP and BNP), which are potent vasoactive hormones with a key role in cardiovascular homeostasis.  NPR1 plays an essential role in the regulation of endothelial cell senescence and vascular aging.  Upon activation by ANP or BNP, NPR1 stimulates the production of cyclic guanosine monophosphate (cGMP) that promotes vascular tone and volume homeostasis by activation of protein kinase cascade, thereby controlling blood pressure and maintaining cardiovascular homeostasis.
