Description

LAMP2 (Lysosome-associated membrane glycoprotein 2) or CD107b is a single pass type I transmembrane glycoprotein belonging to the LAMP family. LAMP2 contains 2 extracellular lumenal domains, a transmembrane and a cytoplasmic domain.  LAMP2 may present carbohydrate ligands to selectins and is implicated in tumor cell metastasis.  LAMP2 may also function in the protection, maintenance, and adhesion of the lysosome. LAMP2 plays an important role in chaperone-mediated autophagy, a process that mediates lysosomal degradation of proteins in response to various stresses and as part of the normal turnover of proteins with a long biological half-live.  LAMP2 is required for efficient MHCII-mediated presentation of exogenous antigens via its function in lysosomal protein degradation; antigenic peptides generated by proteases in the endosomal/lysosomal compartment are captured by nascent MHCII subunits.  Mutations in the LAMP2 gene are associated with Danon disease (DAND), a lysosomal glycogen storage disease characterized by the clinical triad of cardiomyopathy, vacuolar myopathy and mental retardation. DAND is often associated with an accumulation of glycogen in muscle and lysosomes.

Construct Details: Full length human LAMP2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
