VSTM1 (V-set and transmembrane domain-containing protein 1) or SIRL1 (Signal inhibitory receptor on leukocytes-1) is a single pass type I transmembrane protein belonging to the Immunoglobulin superfamily (IgSF).  VSTM1 contains a 119 aa extracellular region (ECD) with 1 V-type Ig­-like (IgV) domain, a transmembrane (TM) domain and a 80 aa cytoplasmic tail with 2 copies of immunoreceptor tyrosine-based inhibitor motif (ITIM). The ITIM is involved in modulation of cellular responses. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases. Both motives are required for full inhibition of FCER1A-mediated degranulation.  VSTM1 is expressed on myeloid (neutrophils, eosinophils and monocytes) but not on lymphoid cells.  VSTM1 may act as an inhibitory immune receptor involved in the regulation of phagocytes.  The Isoform 2 of VSTM1 (v2) which lacks the TM domain behaves as a cytokine, promoting IL17A secretion by CD4+ T-cells, and differentiation and activation of IL17 producing helper T-cells (TH17).

Construct Details: Full length human VSTM1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
