ALK receptor tyrosine kinase (RTK), is a single-pass type I transmembrane glycoprotein that belongs to the insulin receptor (InsR)subfamily of the tyrosine protein kinase family. ALK comprises a 1020-aa large extracellular domain (ECD), a transmembrane region (TM), and a 561-aa intracellular kinase domain. ALK is a neuronal RTK that is expressed in specific regions of the central and peripheral nervous systems and plays an important role in the genesis and differentiation of the nervous system. ALK acts as a receptor for ligands pleiotrophin (PTN), a secreted growth factor, and midkine (MDK), a PTN-related factor, thus participating in PTN and MDK signal transduction, leading to the activation of the mitogen-activated protein kinase (MAPK) pathway.  The MAPK pathway is important for the anti-apoptotic signaling of PTN and regulation of cell proliferation.  ALK drives NF-kappa-B activation, probably through IRS1 and the activation of the AKT serine/threonine kinase. The ALK gene has been found to be rearranged, mutated, or amplified in many types of cancer including anaplastic large cell lymphomas, neuroblastoma, and non-small cell lung cancer (NSCLC). The chromosomal rearrangements are the most common genetic alterations in the ALK gene, which result in creation of multiple fusion genes in tumorigenesis, including ALK/EML4, ALK/RANBP2, ALK/ATIC, ALK/TFG, ALK/NPM1, ALK/SQSTM1, ALK/KIF5B, ALK/CLTC, ALK/TPM4, and ALK/MSN.

Construct Details: Full length human ALK/CD246 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
