ATP1B4 (ATPase Na+/K+ transporting subunit beta 4) is a single-pass type II transmembrane glycoprotein beloning to the family of Na+/K+ and H+/K+ ATPases beta chain proteins. Na+/K+ -ATPase is an integral membrane protein responsible for establishing and maintaining the electrochemical gradients of Na and K ions across the plasma membrane. These gradients are essential for osmoregulation, for sodium-coupled transport of a variety of organic and inorganic molecules, and for electrical excitability of nerve and muscle. This ATPase is composed of three subunits: a large catalytic subunit alpha (catalytic), a smaller glycoprotein subunit beta and gamma. The beta subunit regulates, through assembly of alpha/beta heterodimers, the number of sodium pumps transported to the plasma membrane. ATP1B4 may act as a transcriptional co-regulator during muscle development through its interaction with SNW1. ATP1B4 has lost its ancestral function as a Na,K-ATPase beta-subunit as it does not associate with known Na,K-ATPase alpha-subunits. ATP1B4 interacts with the nuclear transcriptional co-regulator SKIP involved in the regulation of TGF-beta signaling.

Construct Details: Full length human ATP1B4 is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
