ASGR2 is a subunit of the asialoglycoprotein receptor (ASGR/ASGPR). ASGR2 is a single-pass type II transmembrane protein that belongs to a the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily. ASGR is a hetero-oligomeric protein composed of major and minor subunits encoded by different genes. ASGR2 is the more abundant major subunit. ASGR2 plays a critical role in serum glycoprotein homeostasis by mediating the endocytosis and lysosomal degradation of glycoproteins. ASGR2 mediates the endocytosis of plasma glycoproteins to which the terminal sialic acid residue on their complex carbohydrate moieties has been removed. ASGR2 recognizes terminal galactose and N-acetylgalactosamine units. After ligand binding to the receptor, the resulting complex is internalized and transported to a sorting organelle, where receptor and ligand are disassociated. The receptor then returns to the cell membrane surface. Calcium is required for ligand binding. ASGR2 may facilitate hepatic infection by multiple viruses including hepatitis B, and is also a target for liver-specific drug delivery.

Construct Details: Full length human ASGR2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
