IgSF8 or CD316 is a 613 amino acid (aa) single pass type I transmembrane glycoprotein that belongs to the EWI subfamily of the Immunoglobulin superfamily (IgSF). IgSF8 contains an N-terminal signal peptide, a 552-aa extracellular domain (ECD) with 4 Ig-like C2-type domains, an EWI (Glu-Trp-Ile)-motif and 3 potential glycosylation sites, a transmembrane domain (TM), and a 13-aa short cytoplasmic tail. IgSF8 interacts with the tetraspanins CD9 and CD81 and may regulate their role in certain cellular functions including cell migration, oocytes fertilization and viral infection. IGSF8 may regulate proliferation and differentiation of keratinocytes. IgSF8 may also function as a tumor suppressor by inhibiting the proliferation of certain cancers. IgSF8 may be a negative regulator of cell motility: suppresses T-cell mobility coordinately with CD81, associates with CD82 to suppress prostate cancer cell migration, regulates epidermoid cell re-aggregation and motility. IgSF8 may also play a role on integrin-dependent morphology and motility functions. IgSF8 may participate in the regulation of neurite outgrowth and maintenance of the neural network in the adult brain.

Construct Details: Full length human IgSF8 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
