JAM4 (Junctional adhesion molecule 4) or IgSF5 is a 407 amino acid (aa) single pass type I transmembrane glycoprotein that belongs to the Immunoglobulin superfamily (IgSF). JAM4 contains an N-terminal signal peptide, a 266-aa extracellular domain (ECD) with 2 Ig-like V-type domains and 4 potential glycosylation sites, a transmembrane domain (TM), and a 122-aa cytoplasmic tail. JAM4 provides, together with MAGI1, an adhesion machinery at tight junctions, which may regulate the permeability of kidney glomerulus and small intestinal epithelial cells. JAM4 mediates calcium-independent homophilic cell adhesion. In testis, it may function as a cell adhesion molecule rather than a tight-junction protein. JAM4may participate in the adhesion between spermatogonia-spermatogonia, spermatogonia-Sertoli cells, and Sertoli cells-Sertoli cells.

Construct Details: Full length human JAM4 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
