TWEAK (TNF-related weak inducer of apoptosis) is a member of the tumor necrosis factor superfamily (TNFSF12). TWAEK/TNFSF12 is a single-pass type II transmebrane glycoprotein consisting of a 21 amino acid (aa) cytoplasmic domain, a transmembrane domain and a 207 aa extracellular domain (ECD). TWAEK/TNFSF12 may be released into the extracellular fluid, probably by cleavage form the cell surface. TWAEK/TNFSF12 is a ligand for the FN14/TWEAKR receptor. TWAEK/TNFSF12 has overlapping signaling functions with TNF, but displays a much wider tissue distribution. TWAEK/TNFSF12, which exists in both membrane-bound and secreted forms, can induce apoptosis via multiple pathways of cell death in a cell type-specific manner. TWAEK/TNFSF12 is also found to promote proliferation and migration of endothelial cells, and thus acts as a regulator of angiogenesis.

Construct Details: Full length human TWEAK/TNFSF12 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter -/+ an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

 

Vector Type: pLTC or pLTC-Marker (lentiviral expression vector containing a heterologous CMV promoter -/+ a selection marker, see the vector map above)

 

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
