LIGHT, also known as TNFSF14, HVEML and CD258, is a cytokine that belongs to the tumor necrosis factor (TNF) superfamily. LIGHT is a single-pass type II transmebrane glycoprotein consisting of a 37 amino acid (aa) cytoplasmic domain, a 21-aa transmembrane domain (TM) and a 182 aa extracellular domain (ECD). LIGHT is expressed by activated T lymphocytes, monocytes, granulocytes, spleen cells, and immature dendritic cells. LIGHT is known to bind at lest 2 TNF receptor superfamily (TNFRSF) members: HVEM, which is predominantly expressed by T cells, and LTβR (lymphotoxin β receptor), which is expressed by stromal cells and nonlymphoid hematopoietic cells. LIGHT also binds to a soluble TNFRSF member, DcR3 (decoy receptor 3), which can modulate the function of LIGHT. LIGHT functions as a co-stimulatory factor for the activation of lymphoid cells and as a deterrent to infection by herpesvirus. LIGHT activates NF-kappaB, stimulates the proliferation of T cells, and triggers apoptosis of various tumor cells. LIGHT is also reported to prevent TNF-mediated apoptosis in primary hepatocyte. LIGHT has been shown to suppress tumor formation through the up-regulation of intercellular adhesion molecule 1 and the increase of lymphocyte adhesion to cancer cells. LIGHT is thus a promising target for cancer treatment.

Construct Details: Full length human LIGHT gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.
