Description

NPR2 (natriuretic peptide receptor 1), also known as GC-B, ANPRB and GUCY2B, is a single pass type I transmembrane glycoprotein that belongs to the adenylyl cyclase class-4/guanylyl cyclase family. Guanylyl cyclases catalyze the production of cGMP from GTP and exist as soluble and membrane forms.  The membrane guanylyl cyclases, often termed guanylyl cyclases A through F, are a family of cell-surface receptors with a similar topographic structure: an extracellular domain (ECD) for ligand-binding, a transmembrane (TM) domain, and an intracellular region that contains a protein kinase-like domain, a helical hinge region involved in oligomerization, and a carboxyl-terminal guanylyl cyclase catalytic domain. NPR2 is a membrane-bound guanylate cyclase that serves as the primary receptor for the C-type natriuretic peptide NPPC/CNP hormone. NPR2 may play a role in the regulation of skeletal growth. The mutations in the NPR2 gene are associated with AMD1 (acromesomelic dysplasia 1), a skeletal disorder characterized by short stature, very short limbs and hand/foot malformations, and ECDM (epiphyseal chondrodysplasia, miura type), an overgrowth syndrome characterized by tall stature, long hands and feet.
