CD123, also known as IL3RA/IL3R (Interleukin-3 receptor subunit alpha), is a single pass, type I transmembrane glycoprotein of the type I cytokine receptor family, type 5 subfamily.  CD123/IL3RA consists of an extracellular region and a juxtamembrane WSXWS motif, a transmembrane domain, and a cytoplasmic tail with the box 1 motif.  The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding, while the box 1 motif is required for JAK interaction/activation. CD123/IL3RA is a component of the receptor complex specific for IL3. The IL3 receptor complex is a heterodimer of CD123/IL3RA and the common signal transducing subunit CD131/CSF2RB shared by the receptors for IL3, colony stimulating factor 2 (CSF2/GM-CSF), and IL5. The binding of CD123/IL3RA to IL3 depends on the beta subunit CD131. The beta subunit CD131 is activated by the ligand binding, and is required for the biological activities of IL3. This gene and the gene encoding the colony stimulating factor 2 receptor alpha chain (CSF2RA/CD116) form a cytokine receptor gene cluster in a X-Y pseudoautosomal region on chromosomes X or Y.

Construct Details: The recombinant human CD123 ECD protein is expressed as a 306 amino acid protein consisting of Thr19 - Ala303 region of CD123 and a C-terminal His-tag. It contains 6 potential sites for N-linked glycosylation.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
