Human TLR9/CD289 Gene, Full-length cDNA in Lentivector, Endotoxin-free DNA
TLR9 is a single-pass type I transmembrane glycoprotein belonging to the Toll-like receptor (TLR) family. The TLR family members play a fundamental role in pathogen recognition and activation of innate immunity. They serve as pattern recognition receptors for microbial pathogens. There are at least 11 mouse and 10 human TLR members. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. TLRs contain a large number of leucinerich repeats (LRRs) and a cytoplasmic tail with a Toll/IL1 receptor (TIR) domain. TLRs recognize pathogen-associated molecular patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. TLR9 contains 26 LRR repeats in the extracellular region. The various TLRs exhibit different patterns of expression. TLR9 is preferentially expressed in immune cell rich tissues, such as spleen, lymph node, bone marrow and peripheral blood leukocytes. Studies in mice and human indicate that this receptor mediates cellular response to unmethylated CpG dinucleotides in bacterial DNA to mount an innate immune response.

Construct Details: Full length human TLR9/CD289 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter +/- an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.
