Mouse Nanog Full-length Gene in Lentivector, Endotoxin-free DNA
Nanog is a transcription factor involved in inner cell mass and embryonic stem (ES) cells proliferation and self-renewal. It contains 1 conserved homeobox DNA-binding domain at the N-terminal region and is the founding member of the Nanog homeobox family. Nanog binds optimally to the DNA consensus sequence 5'-TAAT[G/T][G/T]-3' or 5'-[C/G][G/A][C/G]C[G/C]ATTAN[G/C]-3'. Nanog is expressed in embryonic stem (ES) and carcinoma (EC) cells. Nanog may be useful in the immunohistochemical diagnosis of tumors. NANOG is thought to be a key factor in maintaining pluripotency and function in concert with other transcription factors such as Oct-4 (POU5F1) and SOX2 to establish ES cell identity bearing the ability to become virtually any cell of any of the 3 germ layers (endoderm, ectoderm, mesoderm). Overexpression of Nanog in mouse embryonic stem cells causes them to self-renew in the absence of Leukemia inhibitory factor (LIF). Overexpression of Nanog in human embryonic stem cells enables their propagation for multiple passages during which the cells remain pluripotent. Loss of Nanog function causes differentiation of mouse or human embryonic stem cells into other cell types.

Construct Details: Full length mouse Nanog gene is subcloned into the Lentiviral expression vector pLTE with an upstream EF1α promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple selection markers are available.
