Human DcR2 Full-length Gene in Lentivector, Endotoxin-free DNA
DcR2 ( decoy receptor 2), also known as CD264, TRAILR4, and TRUNDD, is a Single-pass type I transmembrane protein of the TNF receptor superfamily (TNFRSF10D). DcR2 contains an extracellular TRAIL-binding domain with 3 TNFR-cysteine repeats, a transmembrane domain, and a truncated cytoplasmic death domain. Therefore DcR2 does not induce apoptosis, and may play an inhibitory role in TRAIL-induced cell apoptosis. DcR2 is widely expressed, in particular in fetal kidney, lung and liver, and in adult testis and liver. It is also expressed in peripheral blood leukocytes, colon and small intestine, ovary, prostate, thymus, spleen, pancreas, kidney, lung, placenta and heart. Aberrant methylation of DcR2 is frequent in multiple tumor types. Overexpression of TRAIL R4 can protect cells bearing TRAILR1 and/or TRAILR2 from TRAILmediated apoptosis. The human soluble DcR2-Fc chimera neutralizes the ability of TRAIL to induce apoptosis.
Gene Symbol: DCR2; CD264; TRUNDD; TRAILR4; TRAIL-R4; UNQ251/PRO288
NCBI Gene ID: 8793
Uniprot Entry: Q9UBN6
Construct Details: Full length human DcR2/CD264 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 1161 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
