Description

NCR2, also known as LY95, NKp44 and CD336, is a single-pass type I transmembrane glycoprotein that contains an extracellular Immunoglobulin (Ig)-like domains and a short cytoplasmic tail.  NCR2/NKp44/CD336 together with NCR1/NKp36/CD335 and NCR3/NKp30/CD337 are activating receptors collectively termed the natural cytotoxicity receptors (NCRs).  All these receptors lack significant sequence homology to one another. They are expressed almost exclusively by NK cells and are primarily responsible for triggering NK cell-mediated lytic activities of human tumor cells.  Blocking any of these three NCRs individually resulted in partial inhibition of tumor cell lysis, but nearly complete inhibition of lysis was observed if all three receptors were blocked simultaneously.  NCR2 may contribute to the increased efficiency of activated natural killer (NK) cells to mediate tumor cell lysis.

Gene Symbol: NCR2; LY95; CD336; NKP44; NK-p44; dJ149M18.1; NCTR2

NCBI Gene ID: 9436

Uniprot Entry: O95944

Construct Details: Full length human NCR2/CD336/NKp44 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Gene Insert Size: 831 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
