Description

Nectin2 or CD112 is a single pass type I transmembrane glycoprotein that belongs to the Nectin family of the Immunoglobulin superfamily (IgSF).  Most Nectin family members function as cell adhesion molecules (CAMs) located on the cell surface and involved with the binding with other cells or with the extracellular matrix (ECM). Nectin2 contains 1 Ig-like V-type domain and 2 Ig-like C2-type domains in the extracellular region that promotes cell-cell contacts by forming homophilic or heterophilic trans-dimers. Heterophilic interactions have been detected between NECTIN2 and NECTIN3. It binds with low affinity to TIGIT. Nectin2 may modulate T-cell signaling, acting as either a costimulator of T-cell function, or a coinhibitor, depending on the receptor it binds to. Upon binding to CD226, Nectin 2 stimulates T-cell proliferation and cytokine production, including that of IL2, IL5, IL10, IL13, and IFNG. Upon interaction with PVRIG, Nectin 2 inhibits T-cell proliferation. Nectin2 functions as a receptor for herpes simplex virus 1 (HHV-1) mutant Rid1, herpes simplex virus 1 (HHV-2) and pseudorabies virus (PRV) envelope glycoprotein D. Variations in the Nectin2 gene are associated with differences in the severity of multiple sclerosis (MS).

Construct Details: The recombinant human Nectin2/CD112 ECD protein is expressed as a 340-amino acid active form consisting of Gln32 -  Leu360 region of Nectin2 isoform alpha (UniProt #Q92692-2) and a C-terminal His-tag. Nectin2 ECD contains 2 potential N-linked glycosylation sites.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
