Human Tie2 Full-length Gene in Lentivector, Endotoxin-free DNA
Tie1 (Tyrosine kinase with Ig and EGF homology domains 1) and Tie2 comprise a receptor tyrosine kinase (RTK) subfamily with unique structural features: two immunoglobulinlike (Ig) domains flanking three epidermal growth factor (EGF)like domains and followed by three fibronectin type IIIlike repeats in the extracellular region and a split tyrosine kinase domain in the cytoplasmic region. Tie1 and Tie2 are expressed primarily on endothelial and hematopoietic progenitor cells and play critical roles in angiogenesis, vasculogenesis, and hematopoiesis. Tie2 is a receptor for the angiopoietin (ANG) family: ANG1, ANG2, and ANG3 (mouse)/ANG4 (human). Tie1 may also act as an ANG receptor, possibly in complex with Tie2. Tie2 regulates angiogenesis, endothelial cell survival, proliferation, migration, adhesion and cell spreading as well as the reorganization of the actin cytoskeleton and maintenance of vascular quiescence. Tie2 is required for normal angiogenesis and heart development during embryogenesis, and for post-natal hematopoiesis. Tie2 may activate or inhibit angiogenesis, depending on the context. Tie2 may have anti-inflammatory effects by preventing the leakage of proinflammatory plasma proteins and leukocytes from blood vessels. Tie2 can be proteolytic processing leads to the shedding of the extracellular domain (soluble Tie2 or sTie2). Tie2 may play a role in a range of diseases with a vascular component, including neovascularization of tumors, psoriasis and inflammation. Mutations in the Tie2 gene are associated with dominantly inherited venous malformations (VMCM).

Construct Details: Full length human Tie2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.
