Description

BTN2A2 (Butyrophilin subfamily 2 member A2) is a single pass type I transmembrane protein belonging to the BTN/MOG family of the immunoglobulin (Ig) superfamily (IgSF). Butyrophilin (BTN) is the major protein associated with fat droplets in the milk.  The BTN genes are a group of major histocompatibility complex (MHC)-associated genes that encode type I transmembrane proteins with 2 extracellular immunoglobulin (Ig) domains and an intracellular B30.2/SPRY (PRYSPRY) domain. Three subfamilies of human BTN genes are located in the MHC class I region: the single-copy BTN1A1 gene and the BTN2 (e.g., BTN2A1) and BTN3 (e.g., BNT3A1) genes, which have undergone tandem duplication, resulting in 3 copies of each.  BTN2A2 contains 1 Ig-like V-type (IgV) and 1 Ig-like C2-type (IgC2) domains in the extracelluar region (ECD). BTN2A2 inhibits the proliferation of CD4 and CD8 T-cells activated by anti-CD3 antibodies, T-cell metabolism and IL2 and IFN-Gamma secretion. BTN2A2 may also be involved in lipid, fatty-acid and sterol metabolism.

Gene Symbol: BTN2A2; BTF2; BT2.2; BTN2.2

NCBI Gene ID: 10385

Uniprot Entry: Q8WVV5

Construct Details: Full length human BTN2A2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter +/- an antibiotics selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Gene Insert Size: 1572 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
