Human SRPX2/BPP/CBPS/SRPUL/RESDX Gene, Full-length cDNA in Lentivector, Endotoxin-free DNA
SRPX2 (sushi repeat containing protein, X-linked 2) is a secreted glycoprotein with 3 conserved Sushi (CCP/SCR) domains in the extracellular region. Sushi is a motif of ~60 amino acids characterized by a framework of 4 conserved half-cystine residues (1-3 and 2-4 disulfide-bonded) and several other highly conserved residues including proline, tryptophan, tyrosine/phenylalanine and cysteine. This type of structure, also designated SCR (Short Consensus Repeat), is predominantly present in proteins associated with the complement system. SRPX2 acts as a ligand for the urokinase plasminogen activator surface receptor. SPRX2 plays a role in angiogenesis by inducing endothelial cell migration and the formation of vascular network. It is involved in cellular migration and adhesion. SRPX2 increases the phosphorylation levels of FAK and interacts with and increases the mitogenic activity of HGF. SRPX2 promotes synapse formation and may have a role in the perisylvian region, critical for language and cognitive development. SRPX2 may play a role in the development of speech and language centers in the brain Mutations in the SRPX2 gene are the cause of bilateral perisylvian polymicrogyria, rolandic epilepsy with speech dyspraxia and mental retardation X-linked (RESDX).
Gene Symbol: SRPX2; BPP; CBPS; PMGX; RESDX; SRPUL
NCBI Gene ID: 27286
Uniprot Entry: O60687
Construct Details: Full length human SRPX2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 1398 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
