Human LYPD6B/CT116/LYPD7 Gene, Full-length cDNA in Lentivector, Endotoxin-free DNA
LYPD6B (Ly6/PLAUR domain-containing protein 6B) is a glycosyl-phosphatidylinositol (GPI)-linked cell surface glycoprotein. A variety of GPI-linked surface glycoproteins are composed of one or more copies of a conserved domain of ~100 amino-acid residues termed u-PAR/Ly-6 motif that was originally found in u-PAR (Urokinase plasminogen activator surface receptor) and Ly-6/CD59. The u-PAR/Ly-6 (UL) motif has 10 conserved cysteines with a defined disulfide-bonding pattern. LYPD6B contains 1 copy of u-PAR/Ly6 motifs in the extracellular domain (ECD). LYPD6B acts as a modulator of nicotinic acetylcholine receptors (nAChRs) function in the brain and inhibits nicotine-induced Ca2+ influx through nAChRs. LYPD6B acts on nAChRs in a subtype- and stoichiometry-dependent manner. It modulates specifally alpha-33:beta-42 nAChRs by enhancing the sensitivity to ACh, decreasing ACh-induced maximal current response and increasing the rate of desensitization to ACh. LYPD6B has no effect on alpha-7 homomeric nAChRs. It also modulates alpha-32:alpha-5:beta-42 nAChRs in the context of CHRNA5/alpha-5 variant Asn-398 but not its wild-type sequence.

Construct Details: Full length human LYPD6B gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
