IFITM1 (interferon-induced transmembrane protein 1) also known as CD225 and DSPA2a (dispanin subfamily A member 2a) is a single-pass transmembrane protein belonging to the CD225/Dispanin family. IFTIM1 is an interferon-induced antiviral protein which inhibits the entry of viruses to the host cell cytoplasm, permitting endocytosis, but preventing subsequent viral fusion and release of viral contents into the cytosol. It is active against multiple viruses, including influenza A virus, SARS coronavirus (SARS-CoV), Marburg virus (MARV), Ebola virus (EBOV), Dengue virus (DNV), West Nile virus (WNV), human immunodeficiency virus type 1 (HIV-1) and hepatitis C virus (HCV). IFITM1 can inhibit: influenza virus hemagglutinin protein-mediated viral entry, MARV and EBOV GP1,2-mediated viral entry and SARS-CoV S protein-mediated viral entry. Palmitoylation on membrane-proximal cysteines controls clustering in membrane compartments and antiviral activity against influenza virus.  IFITM1 ilso implicated in cell adhesion and control of cell growth and migration. IFITM1 plays a key role in the antiproliferative action of IFN-gamma either by inhibiting the ERK activation or by arresting cell growth in G1 phase in a p53-dependent manner. It aslo acts as a positive regulator of osteoblast differentiation.  IFITM1 is expressed in bone and glioma cell lines.  Its expression levels are greatly elevated in colon cancer, cervical cancer, esophageal cancer and ovarian cancer.

Construct Details: Full length human IFITM1/CD225 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
