S1PR1 (sphingosine-1-phosphate receptor 1), also known as S1P1, EDG1 and CD363, is a 7-transmembrane protein of the G protein-coupled receptor 1 (GPCR1) family. S1PR1 is highly expressed in endothelial cells and binds the ligand sphingosine-1-phosphate (S1P) with high affinity and high specificity. S1P is a bioactive lysophospholipid that elicits diverse physiological effect on most types of cells and tissues including proliferation, apoptosis, motility, and neurite retraction. S1P acts both intracellularly as a second messenger and extracellularly as a receptor ligand. S1P and the structurally related lysolipid mediator lysophosphatidic acid (LPA) signal cells through a set of GPCR known as EDG receptors. Some EDG receptors (e.g., EDG1) are S1P receptors; others (e.g., EDG2) are LPA receptors. S1PR1 is involved in the processes that regulate the differentiation of endothelial cells. Activation of S1PR1 leads to the activation of RAC1, SRC, PTK2/FAK1 and MAP kinases. S1PR1 plays an important role in cell migration, probably via its role in the reorganization of the actin cytoskeleton and the formation of lamellipodia in response to stimuli that increase the activity of the sphingosine kinase SPHK1. S1PR1 is required for normal chemotaxis toward S1P and for normal embryonic heart development and normal cardiac morphogenesis. S1PR1 plays an important role in the regulation of sprouting angiogenesis and vascular maturation. It is also required for normal egress of mature T-cells from the thymus into the blood stream and into peripheral lymphoid organs. S1PR1 plays a role in the migration of osteoclast precursor cells, the regulation of bone mineralization and bone homeostasis.

Construct Details: Full length human S1PR1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
