Human CD300f/CLM1/IREM1/IgSF13 Gene, Full-length cDNA in Lentivector, Endotoxin-free DNA
CD300f or CLM1 (CMRF35-like molecule 1) is a 290 amino acid (aa) single pass type I transmembrane glycoprotein that belongs to the CD300 family of the Immunoglobulin superfamily (IgSF). The CD300 family members contain a single Ig-like V-type domain and are found on leukocytes involved in immune response signaling pathways. CD300f contains an N-terminal signal peptide, a 137-aa extracellular domain (ECD) with 1 Ig-like V-type domain and 1 potential glycosylation sites, a transmembrane domain (TM), and a 113-aa cytoplasmic tail. CD300f is expressed on the surface of mature hematopoietic cells of monocytes. CD300f positively regulates the phagocytosis of apoptotic cells (efferocytosis). CD300f also plays an important role in immune homeostasis, by promoting macrophage-mediated efferocytosis and by inhibiting dendritic cell-mediated efferocytosis. CD300f negatively regulates Fc epsilon receptor-dependent mast cell activation and allergic responses via binding to ceramide and sphingomyelin. CD300f may act as a co-receptor for interleukin 4 (IL-4) and regulates IL-4 receptor alpha (IL4Ra)-mediated responses by augmenting IL-4- and IL-13-induced signaling. CD300f negatively regulate the Toll-like receptor (TLR) signaling. CD300f inhibits osteoclast formation and induces macrophage cell death upon engagement
Gene Symbol: CD300f; CLM1; NKIR; CLM-1; IREM1; LMIR3; CD300f; IREM-1; IgSF13
NCBI Gene ID: 146722
Uniprot Entry: Q8TDQ1
Construct Details: Full length human CD300f gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 873 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
