Human Nectin4/PRR4/PVRL4 Gene, Full-length cDNA Clone in Lentivector, Endotoxin-free DNA
Nectin4 is a single pass type I transmembrane glycoprotein that belongs to the Nectin family of the Immunoglobulin superfamily (IgSF). Most Nectin family members function as cell adhesion molecules (CAMs) located on the cell surface and involved with the binding with other cells or with the extracellular matrix (ECM). Nectin4 contains 1 Ig-like V-type domain and 2 Ig-like C2-type domains in the extracellular region that promotes cell-cell contacts by forming homophilic or heterophilic trans-dimers. Heterophilic interactions have been detected between Nectin4 and Nectin1. A soluble form of Nectin4 can be produced by proteolytic cleavage at the cell surface by the metalloproteinase ADAM17/TACE. The secreted Nectin4 is found in both breast tumor cell lines and breast tumor patients. Nectin4 acts as a receptor for measles virus but not for alpha-herpesvirus entry into cells. Mutations in the Nectin4 gene are the cause of ectodermal dysplasia-syndactyly syndrome type 1, an autosomal recessive disorder.
Gene Symbol: Nectin4; LNIR; PRR4; EDSS1; PVRL4; nectin-4
NCBI Gene ID: 81607
Uniprot Entry: Q96NY8
Construct Details: Full length human NECTIN4 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter -/+ an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.
Vector Type: pLTC or pLTC-Marker (lentiviral expression vector containing a heterologous CMV promoter -/+ a selection marker, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 1533 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CGGGAAGCAATAGCATGATA-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
