Human Nectin2/CD112/PRR2/PVRL2 Gene, Full-length cDNA Clone in Lentivector, Endotoxin-free DNA
Nectin2 or CD112 is a single pass type I transmembrane glycoprotein that belongs to the Nectin family of the Immunoglobulin superfamily (IgSF). Most Nectin family members function as cell adhesion molecules (CAMs) located on the cell surface and involved with the binding with other cells or with the extracellular matrix (ECM). Nectin2 contains 1 Ig-like V-type domain and 2 Ig-like C2-type domains in the extracellular region that promotes cell-cell contacts by forming homophilic or heterophilic trans-dimers. Heterophilic interactions have been detected between NECTIN2 and NECTIN3. It binds with low affinity to TIGIT. Nectin2 may modulate T-cell signaling, acting as either a costimulator of T-cell function, or a coinhibitor, depending on the receptor it binds to. Upon binding to CD226, Nectin 2 stimulates T-cell proliferation and cytokine production, including that of IL2, IL5, IL10, IL13, and IFNG. Upon interaction with PVRIG, Nectin 2 inhibits T-cell proliferation. Nectin2 functions as a receptor for herpes simplex virus 1 (HHV-1) mutant Rid1, herpes simplex virus 1 (HHV-2) and pseudorabies virus (PRV) envelope glycoprotein D. Variations in the Nectin2 gene are associated with differences in the severity of multiple sclerosis (MS).

Construct Details: Full length human NECTIN2 gene (encoding isoform alpha or Uniprot#Q92692-2) is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter -/+ an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.
