Human CNTFR/CNTFRA Gene, Full-length cDNA Clone in Lentivector, Endotoxin-free DNA
CNTFR or CNTFRa (Ciliary neurotrophic factor receptor subunit alpha) is a GPI-anchored surface glycoprotein of the type I cytokine receptor family, type 3 subfamily. CNTFR consists of an extracellular region with 1 Ig-like C2-type (immunoglobulin-like) domain, 2 fibronectin type III (FN3) domains and a juxtamembrane WSXWS motif, in addition to a transmembrane (TM) domain, and a cytoplasmic tail. The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding. CNTF is one of the cytokines including IL6, LIF, OSM, CNTF, IL11 and CT1 that utilize the common signaling transducer IL6ST/gp130 for initiating signal transmission. Binding of CNTF to CNTFR recruits the transmembrane components of the co-receptors, IL6ST/gp130 and leukemia inhibitory factor receptor (LIFR), facilitating signal transduction. The CNTF signaling pathway plays a critical role in neuronal cell survival, differentiation and gene expression. Single nucleotide polymorphisms in the CNTFR gene may be associated with variations in muscle strength, as well as early onset of eating disorders.

Construct Details: Full length human CNTFR gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
