Human CD122/IL2RB/IL15RB Gene, Full-length cDNA Clone in Lentivector, Endotoxin-free DNA
CD122, also known as IL2RB (Interleukin-2 receptor subunit beta) and IL15RB, is a single pass, type I transmembrane glycoprotein of the type I cytokine receptor family, type 4 subfamily. CD122 consists of an extracellular region with 1 fibronectin type III (FN3) domains and a juxtamembrane WSXWS motif, a transmembrane domain (TM), and a cytoplasmic tail with the box 1 motif. The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding, while the box 1 motif is required for JAK interaction/activation. IL2RB/CD122 is a component of the receptor complex for IL2 (IL2R), which is a non-covalent dimer of an alpha and a beta subunit. IL2R exists in 3 different forms: a high affinity dimer, an intermediate affinity monomer (beta subunit), and a low affinity monomer (alpha subunit). The high and intermediate affinity forms also associate with a gamma subunit. The beta subunit CD122 is involved in receptor mediated endocytosis and transduces the mitogenic signals of IL2. CD122 interacts with SHB upon interleukin stimulation and with HTLV-1 accessory protein p12I. CD122 is involved in T cell-mediated immune responses.
Gene Symbol: IL2RB; CD122; IL15RB; P70-75
NCBI Gene ID: 3560
Uniprot Entry: P14784
Construct Details: Full length human IL2RB/CD122 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)
Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)
Gene Insert Size: 1656 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:
(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;
(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’
Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification. High fidelity PCR systems should always be used.
