Human CD5/Leu1/Ly1/T1 Protein, ECD (Extracellular Domain), Fc-fusion, Biotinylated, Recombinant
T-cell surface glycoprotein CD5, also known as Leu1, Ly1, and T1, is a single pass type I transmembrane protein belonging to the scavenger receptor superfamily. CD5 contains a 348 amino acid (aa) extracellular domain (ECD) with 3 scavenger receptor cysteinerich (SRCR) domains. Crystal structure of the 3rd ECD of CD5 reveals the fold of a group B SRCR domain. The ECD of CD5 can be cleaved and circulates in the serum. T cell antigen receptor (TCR) positively regulates CD5 expression on developing thymocytes and on peripheral CD4+ T cells. CD5 is known to inhibit TCR signaling and promotes T cell anergy and survival. CD5 signaling negatively regulates the generation of regulatory T cells (Treg) but promotes the development of Th17 cells. CD5 interacts with CD72/Lyb-2 on B cells and with PTPN6/SHP-1 for intracellular signaling. Within the B cell lineage, CD5 is expressed on B1a cells, anergic B cells, and IL10 producing regulatory B cells. Similar to its role on T cells, CD5 negatively regulates signaling through the B cell antigen receptor (BCR) and supports B cell survival, anergy, and tolerance. CD5 is also involved in the cellular entry of hepatitis C virus into T lymphocytes.
