Description

CD127, also known as IL7RA/IL7R (Interleukin-7 receptor subunit alpha), is a single pass, type I transmembrane glycoprotein of the type I cytokine receptor family, type 4 subfamily.  CD127 consists of a extracellular region with 1 fibronectin type III (FN3) domains and a juxtamembrane WSXWS motif, a transmembrane domain, and a cytoplasmic tail with the box 1 motif.  The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding, while the box 1 motif is required for JAK interaction/activation. CD127 is a component of the receptor complex for interleukin-7 and thymic stromal lymphopoietin (TSLP). The IL7 receptor complex is a heterodimer of CD127/IL7R and the common subunit CD132/IL2RG shared by the receptors for interleukins 2, 4, 7, 9, and 15.. The TSLP receptor complex is a heterodimer of TSLPR/CRLF2 and CD127/IL7R. CD127 has been shown to play a critical role in V(D)J recombination during lymphocyte development. Defects in the CD127 gene may be associated with severe combined immunodeficiency (SCID) and multiple sclerosis 3 (MS3).

Construct Details: Full length human CD127/IL7R gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

 

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)

 

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
