Human CD171/L1CAM/NCAML1 Gene, Full-length cDNA Clone in Lentivector, Endotoxin-free DNA
CD171, as known as L1CAM (L1 Cell Adhesion molecule) and NCAML1 (Neural Cell Adhesion Molecule L1), is a single pass type I transmembrane axonal glycoprotein belonging to the L1/Neurofascin/NgCAM family of the Immunoglobulin superfamily (IgSF). CD171 contains a large ~1100-amino acid large extracellular domain (ECD) consisting of 6 Ig-like C2-type motifs and 5 fibronectin-like repeats (type III) linked by a transmembrane (TM) domain to a conserved cytoplasmic tail. CD171 acts as a neural cell adhesion molecule and plays an important role in nervous system development, including neuronal migration, axonal growth and fasciculation, as well as synaptogenesis. In the mature brain, it is critical for the dynamics of neuronal structure and function, including synaptic plasticity. Defects in CD171 are associated with a highly variable clinical spectrum ranging from severe hydrocephalus and prenatal death (HYCX) to a milder phenotype (MASA). Mutations in the CD171 gene cause X-linked neurological syndromes known as CRASH (corpus callosum hypoplasia, retardation, aphasia, spastic paraplegia and hydrocephalus). The features of spasticity are also referred to as complicated spastic paraplegia type 1 (SPG1).

Construct Details: Full length human CD171 gene (encoding UniProt #P32004) is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.
