Cerebral deposition of amyloid beta peptide is an early and critical feature of Alzheimer's disease. Amyloid beta peptide is generated by proteolytic cleavage of amyloid precursor protein (APP) by two proteases, one of which is the aspartic protease encoded by BACE2 gene. BACE2 is a member of the peptidase A1 protein family. It is a type I transmembrane glycoprotein that is found mainly in the Golgi. BACE2 is responsible for the proteolytic processing of the amyloid precursor protein (APP). It cleaves at the N-terminus of the A-beta peptide sequence, between residues 671 and 672 of APP, leads to the generation and extracellular release of beta-cleaved soluble APP, and a corresponding cell-associated C-terminal fragment which is later released by gamma-secretase. BACE2 is present in neurons within the hippocampus, frontal cortex and temporal cortex. It is expressed at low levels in most peripheral tissues and at higher levels in colon, kidney, pancreas, placenta, prostate, stomach and trachea. BACE2 is expressed in the ductal epithelium of both normal and malignant prostate. It is up-regulated in primary breast and colon tumors and liver metastasis. BACE2 interacts ith RTN3 and RTN4 and its activity is inhibited by RTN3/4.

Construct Details: Full length human BACE2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells. It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
